Caribou's distinguishing idea is the guide itself: instead of an all-RNA guide, it uses a hybrid of RNA and DNA, which the company says makes the editing machinery more particular about matching its target exactly. It applies this to off-the-shelf CAR-T cell therapies for blood cancers and autoimmune disease.
Caribou Biosciences was founded in 2011 by Jennifer Doudna, Rachel Haurwitz, Martin Jinek and James Berger. Its chRDNA platform uses hybrid CRISPR RNA-DNA guides with Cas12a, reported to reduce off-target editing. Lead programmes are allogeneic CAR-T candidates including CB-010 (anti-CD19) and CB-011 (anti-BCMA).
